Yes — it's one of the smartest things you can do for the plenum. Bare concrete under a raised floor continuously sheds dust that your underfloor cooling then distributes straight into equipment. Coating the subfloor seals it permanently, turns the plenum into a cleanable surface, and protects the slab from condensation and coolant leaks. It's routinely done during construction or in phases during floor tile maintenance.
